IV. 實驗技術 (Lab techniques)

1.西方點墨法 (Western blot analysis):分析特定蛋白的表現,例如:Connexin 43, eNOS, StAR, Aquporin 7, MAPK, AMPK etc.

2.酵素免疫分析法 (ELISA):分析血液、組織或細胞中特定分子的濃度,例如:Interleukin 1, Interleukin 4, Interleukin 18, TNFa, Ghrelin, luteinizing hormone, and testosterone.

3.Zymography:分析matrix metalprotease (MMP) 活性

  Electrophoresis mobility shift assay (EMSA):分析蛋白與promoter 結合能力

  其他化學分析:

(a)能量代謝分析 (如:Glycogen content, Citrate synthase activity),

(b)氧化壓力及抗氧化能力分析 (MDA, TBARS, and activities of anti-oxidant enzymes, GSH content etc.)

4.組織染色 (Staining):Masson’s trichrome stain (膠原蛋白分佈), Hematoxylin and Eosin stain (病理組織染色), Immunofluorescence stain (免疫螢光染色), Immunohistochemistry stain(免疫組織染色).

5. 細胞培養 (Cell culture)

(a)初代培養 (primary cells):內皮細胞 (endothelial cells), 血管平滑肌細胞,心肌細胞,睪丸萊氏細胞 (Leydig cells of testis), 卵巢顆粒細胞 (Granulosa cells of ovarian follicles), 腎上腺皮質細胞 (Zona glomerulosa and reticularis cell of adrenal gland), 肝臟星狀細胞 (Hepatic Stellate Cell)

(b)細胞株:前列腺癌細胞 (prostate cancer cell line), 腎上腺皮質細胞株 (H295 cells)

6. 動物手術 (Animal Surgery)

(a)留置導管 (Catheterization) 以利動物於清醒無壓力下進行給藥及採集血樣

(b)尾靜脈採血、心室採血、組織或器官摘除
